Synthesis of novel 1α,25-dihydroxyvitamin D3 analogs with unnatural triene system

1α,25-Dihydroxyvitamin D3 [1α,25-(OH)2D3], known also as calcitriol, is the most potent metabolite of vitamin D3. Calcitriol is a hormonal form responsible for calcium and phosphorus homeostasis and it is involved in a wide array of other physiological processes, for example, cell differentiation and proliferation. Such a broad spectrum of activities makes low-calcemic analogs of calcitriol potentially useful in various biomedical applications.
We have designed and synthesized analogs of 1α,25-dihydroxyvitamin D3 with the unnatural triene moiety, “rotated” around C2 axis in comparison with the natural hormone, as well as yet unknown 9-alkylated calcitriol. These newly obtained vitamins are interesting candidates for structure-activity studies. The biological testing of the synthesized compounds is underway.
